You know that Jack was pretty much foaming at the mouth because he was imagining killing Tony! I very much doubt that Tony will survive this season now.
However, I do think that Tony is honestly surprised that there is no cure. I really don't think he realised that, and he's genuinely sorry that Jack is dying.
And yes, Kim has to cure Jack and tell him about his granddaughter!
